In a commercial print setting are both first done with an impo by layout the pages within they will go and print out.

The difference is that with staple binding would be done with the pages placed on their counter part so that when you flip the page that you can see the pages in order. They would take the stack on top of each other and press down and cut. The next is to send it through further and place the staple on the side where you would see the cover first and fold.

Glue binding impo will all on the same sheet folded up and all cut at the same time to make the stack. Then you would take a larger sheet that goes around the booklet and score where you are to fold. The next step is to apply the glue to the edge and let sit till it is done.
